Cell Description:                 Human neuroepithelioma; brain; from metastatic site: supraorbitalarea


Culture medium:                      MEM (Eagle) with Earle«s BSS, sodium bicarbonate, 2mM L-glutamine, 1.0 mM sodium pyruvate, 0,1 mM non-essential amino acids + 10% FBS

Subculture routine:                 Remove medium, rinse with fresh 0,25% trypsin solution, remove trypsin and let the culture sit at room temperature (or at 37”C) until the cells detach (about 10 minutes). Add fresh medium, aspirate  and dispense into new flasks.

Subculture:                             every 6 to 8 days

Split Ratio:                               1:6 to 1:12 is recommended

Morphology:                            Epithelial

Karyotype:                              Hypodiploidy to pseudodiploidy. Abnormalities including double minutes. breaks; large submetacentric, telocentric and small telocentric markers (originator). (P32) Hypodiploid to hyperdiploid and triploid to hypotetraploid with abnormalities including dicentrics, breaks, double minutes (DM), large subtelocentric and small telocentric chromosomes.

Comments:                              This is one of two cell lines (see ATCC HTB-11) of neurogenic origin derived by J. L. Biedler. SK-N-MC was isolated in September of 1971 and was found to have moderate dopamine Š beta Š hydroxylase acivity as well as formaldehyde induced fluorescence indicative of intracellular catecholamines.

Sterility:                                   Tests for Mycoplasma, bacteria and fungi were negative
