Cell Description:                 Human rhabdomyosarcoma

 

Culture medium:                      McCoy's 5a medium with sodium bicarbonate, 2 mM L-glutamine and 1,0 mM sodium pyruvate 90%; fetal bovine serum 10%

Subculture routine:                 Remove medium, rinse with fresh 0.025% trypsin solution, remove trypsin and let the culture sit at room temperature (or at 37¡C) until the cells detach (about 10 minutes). Add fresh medium, aspirate and dispense into new flasks. Subculture every 6 to 8 days.

 

Split Ratio:                               A ratio of 1:6 to 1:10 is recommended

 

Morphology:                            epithelial

Karyotype:                              diploidy and tetraploidy

Tumorigenic:                          yes, in nude mice; forms small cell malignant tumor consistent with embryonal rhabdomyosarcoma; also

                                               in newborn mice treated with anti thymocyte serum

 

Isoenzymes:                          PGM3, 1; PGM1, 1; ES-D, 1; Me-2, 1; AK-1, 1; GLO-1, 1; G6PD, B

Sterility:                                   Tests for Mycoplasma, bacteria and fungi were negative
